Pfizer Says Eczema Drug Shows Significant Skin Clearance

Pfizer Inc. (NYSE:PFE) on Thursday presented detailed Phase 2 results for tilrekimig (PF-07275315) in adults with moderate-to-severe atopic dermatitis.

The ongoing trial met its primary endpoint, showing a statistically significant increase in participants achieving EASI-75 at Week 16 across all evaluated doses compared to placebo.

Strong Efficacy Across Phase 2 Trial Stages

The trial evaluated biologic-naïve adult patients across two distinct stages. Stage 1 tested subcutaneous tilrekimig at 450 mg every two weeks, where 62.5% of patients achieved EASI-75 compared to 19.9% in the placebo group.

Stage 2 evaluated monthly dosing options of 400 mg, 200 mg, or 50 mg. In these monthly cohorts, 58.5%, 61.0%, and 47.8% of patients reached EASI-75, representing absolute improvements of 49.4%, 51.9%, and 38.7% over the 9.1% placebo rate.

Key secondary endpoints measuring clear or almost clear skin (vIGA 0/1) showed 30.3% achievement in Stage 1 versus 11.8% for placebo.

In Stage 2, 26% to 27% of patients across monthly cohorts reached vIGA 0/1, compared to 0% in the placebo group.

An exploratory analysis also demonstrated significant itch reduction, as 42.5% of patients in the 400 mg monthly group and 50.8% in the 200 mg group achieved a 4-point or greater improvement in peak pruritus scores versus 7.2% on placebo.

Novel Mechanism And Safety Results

Tilrekimig is a potential first-in-class trispecific antibody engineered to inhibit type 2 inflammation by targeting IL-4, IL-13, and TSLP. Concurrently blocking TSLP and its downstream effectors provides broader cytokine pathway coverage.

An extended half-life of approximately 37 days supports convenient monthly dosing.

The antibody was well-tolerated with no dose-dependent safety signals.

PFE Price Action: Pfizer shares were down 1.23% at $28.17 at the time of publication on Thursday, according to Benzinga Pro data.
